ABSTRACT

The Goto-Kakizaki (GK) rat is a non-obese experimental model of type 2 diabetes mellitus (T2DM) that allows researchers to monitor diabetes-induced changes without jeopardizing the effects of obesity. This rat strain exhibits notable gastrointestinal features associated with T2DM, such as marked alterations in intestinal morphology, reduced intestinal motility, slow transit, and modified microbiota compared to Wistar rats. The primary treatments for diabetic patients include administration of hypoglycemic agents and insulin, and lifestyle changes. Emerging procedures, including alternative therapies, metabolic surgeries, and modulation of the intestinal microbiota composition, have been shown to improve the diabetic state of GK rats. This review describes the morpho-physiological diabetic-associated features of the gastrointestinal tract (GIT) of GK rats. We also describe promising strategies, e.g., metabolic surgery and modulation of gut microbiota composition, used to target the GIT of this animal model to improve the diabetic state.

ABSTRACT

Abstract Despite their great importance for the food industry, flavorings, in general, raise a number of questions regarding their cytotoxicity, mutagenicity and carcinogenicity, since, in the literature, there are few studies found evaluating the toxicity on the systemic and cellular level, of these chemical compounds. The root meristems of Allium cepa (onion) are widely used for the assessment of toxicity of chemical compounds of interest. Thus, this study aimed to evaluate, in A. cepa meristematic cells, individually and in combination at the cellular level, the toxicity of synthetic Cheese and Cheddar Cheese food flavorings, identical to the natural, at doses of 1.0 and 2.0 mL, at exposure times of 24 and 48 hours. In combination we used 0.5 mL of Cheese flavor associated with 0.5 mL of Cheddar flavor; and 1.0 mL of Cheese flavor associated with 1.0 mL of Cheddar flavor, at exposure times of 24 and 48 hours. For these evaluations, we used groups of five onion bulbs, which were first embedded in distilled water and then transferred to their respective doses. The root tips were collected and fixed in acetic acid (3:1) for 24 hours. The slides were prepared by crushing and were stained with 2% acetic orcein. Cells were analyzed throughout the cell cycle, totaling 5,000 for each control and exposure time. The mitotic indices calculated and cellular aberrations observed were subjected to statistical analysis using the chi-square test (p <0.05). No chromosomal abnormalities nor those of mitotic spindle were observed for the treatments performed. The results, both individually and in combination, showed that the flavorings under study significantly reduced the cell division rate of the test system cells used. Therefore, under the conditions studied, the two flavorings were cytotoxic.

ABSTRACT

Abstract The objective of this study was to evaluate the action of Hymenaea stigonocarpa bark hydroalcoholic extract against a mutagenic compound using A. cepa meristematic root cells as a test system. The treatment groups were: Negative Control (NC) – distilled water; Positive Control (PC) – paracetamol at a concentration of 0.008 mg/mL, Jatoba Control (JC) – aqueous fraction jatobá-do-cerrado at 0.5 or 1.0 or 1.5 mg/mL, and Simultaneous Treatment (ST) - jatobá-do-cerrado aqueous fraction at a concentration of 0.5 or 1.0 or 1.5 mg/mL associated with paracetamol solution at a concentration of 0.008 mg/mL. All groups were analyzed at 24 and 48 h. Five onion bulbs (five replications) were used for each treatment group. The root tips were fixed in Carnoy and slides prepared by the crush technique. Cells were analyzed throughout the cell cycle, totaling 5,000 for each treatment group at each exposure time. Mitotic indices were subjected to statistical analysis using the chi-square test (p<0.05). From the results it was found that the ST group, at the three concentrations, significantly potentiated the antiproliferative effect of the test system cells when compared to PC, NC and TJ at the three concentrations. Furthermore, the three ST concentrations significantly reduced the number of cell aberrations when compared to the number of aberrant cells obtained for the PC, demonstrating antimutagenic action on the A. cepa test system cells.

ABSTRACT

In this study, phytochemical characterization was conducted and the in vitro antibacterial activity of extracts of Ageratum conyzoides L. (whiteweed), Gossypium hirsutum (cotton), Phyllanthus tenellus (shatterstone) and Polygonum hydropiperoides (swamp smartweed) was evaluated against Staphylococcus aureus and Escherichia coli. To assess the antibacterial activity, the agar diffusion method was used. Tests were performed with the extract at alcoholic contents from 0 to 100% (v/v), at 20% proportion (m/v - mass/extractor). Phytochemical tests indicated the presence of reducing sugars, phenolic compounds, flavonoids, tannins, triterpenes and steroids in all four species. The growth of S. aureus cultures was inhibited by all extracts, except for whiteweed extract. The highest inhibitory activity was observed for shatterstone. However, none of the extracts was capable of inhibiting the growth of E. coli strains. Results are promising since three of the four selected plants showed to have antibacterial substances, which stimulates further studies for the isolation and the identification of active principles responsible for this activity, with potential to be used in the pharmaceutical industry.

ABSTRACT

We present data on pulmonary infection rates by parasites in the lizards Tropidurus hispidus Spix, 1825 and T. semitaeniatus (Spix, 1825) living sympatrically in the Chapada do Araripe mountain Range, northeastern Brazil. We found no parasite pulmonary infection in T. semitaeniatus. However, two pulmonary parasite species were found in the T. hispidus hosts, the pentastomid Raillietiella mottae Almeida, Freire and Lopes, 2008 and the nematode Rhabdias sp. Overall prevalence was 5 percent. Prevalence of R. mottae was 2.5 percent and corresponded to only one parasite on each infected host. Prevalence of Rhabdias sp. was 2.5 percent and the range of infection was 1-2 parasites per host. This represents the first record of Rhabdias infecting lizards of the family Tropiduridae in the Neotropical region. Furthermore, we present a comparison of parameters of infection by pulmonary parasites including some recent studies in Brazil.

ABSTRACT

Pulmonary parasitism by pentastomids was examined in two lizard species inhabiting an area of restinga vegetation (coastal sand dunes) situated in the municipality of Mataraca (6° 29' S and 34° 56' W), on the extreme northern coast of Paraíba State, Brazil. A total of 123 lizards were collected, being 75 specimens of Micrablepharus maximiliani (Gymnophtalmidae) and 48 specimens of Cnemidophorus ocellifer (Teiidae). Only a single species of Pentastomida (Raillietiella mottae) was found parasitizing three females M. maximiliani, with a prevalence of 4 percent and an average infection intensity of 2.3 ± 1.3 (range 1-5). The infection rate by pentastomids encountered in the present study was similar to that seen with other species of restinga lizards. Raillietiella mottae is a generalist parasite species that is probably transmitted by common and widely distributed insects making up part of the diet of many insectivorous lizard species from northeastern Brazil.

ABSTRACT

The rates of pentastomid infections in the syntopic lizards Hemidactylus mabouia and Tropidurus hispidus inhabiting buildings at the Federal University of Paraiba, in João Pessoa, NE Brazil were examined. A total of 30 specimens of Hemidactylus mabouia and 55 specimens of Tropidurus hispidus were examined. The animals were sacrificed by freezing and then fixed and conserved in 70 percent alcohol. Analyses of the respiratory tract of Hemidactylus mabouia demonstrated that these lizards were infected by Raillietiella frenatus at a rate of 20 percent (6/30) with an average infection intensity of 1.33 ± 0.21, 1-2. None of the specimens of T. hispidus analyzed were infected. Hemidactylus mabouia and R. frenatus are of African origin and it is possible that there are specific relationships, or preferences, between these two species.

ABSTRACT

Pentastomids can infect the respiratory tract of lizards, causing their death and as a result influencing the population size of hosts. Despite this, studies on rates of pulmonary infection of Brazilian lizards, including those living in Caatinga ecosystems of northeastern Brazil are scarce. Active collections of lizards were performed from October to December 2004 in an area of Caatinga of the Estação Experimental de São João do Cariri - EESJC (07º 25' S and 36º 30' W), located in the state of Paraíba, Northeast of Brazil. Forty-five lizards inhabiting granite outcrops in an area of Caatinga were captured, belonging to the following species: Tropidurus hispidus (Spix, 1825) (18 individuals), T. semitaeniatus (Spix, 1825) (15 individuals), Phyllopezus periosus Rodrigues, 1986 (6 individuals), and P. pollicaris (Spix, 1825) (6 individuals). Laboratory examination revealed that all species had some degree of pulmonary infection caused by Raillietiella mottae. The highest rates of prevalence (66.7 percent) and mean intensity of infection (5.25 ± 2.01, range of 2-11) were observed in P. periosus. The results obtained in this study show that lizards of the Brazilian semi-arid region are infected by a generalist species of pentastomid. The most likely cause for such pattern is the similarity in lizards' diets (ants and termites). It is particularly noteworthy that T. semitaeniatus, P. periosus, and P. pollicaris represent new host records for R. mottae.

ABSTRACT

Third molar extraction is a common procedure frequently accompanied by moderate or severe pain, and involves sufficient numbers of patients to make studies relatively easy to perform. The aim of the present study was to determine the efficacy and safety of the therapeutic combination of 10 mg piroxicam, 1 mg dexamethasone, 35 mg orphenadrine citrate, and 2.5 mg cyanocobalamin (Rheumazin®) when compared with 20 mg piroxicam alone (Feldene®) in mandibular third molar surgery. Eighty patients scheduled for removal of the third molar were included in this randomized and double-blind study. They received (vo) Rheumazin or Feldene 30 min after tooth extraction and once daily for 4 consecutive days. Pain was determined by a visual analogue scale and by the need for escape analgesia (paracetamol). Facial swelling was evaluated with a measuring tape and adverse effects and patient satisfaction were recorded. There was no statistically significant difference in facial swelling between Rheumazin and Feldene (control group). Both drugs were equally effective in the control of pain, with Rheumazin displaying less adverse effects than Feldene. Therefore, Rheumazin appears to provide a better risk/benefit ratio in the mandibular molar surgery. Since the side effects resulting from nonsteroidal anti-inflammatory drug administration are a severe limitation to the routine use of these drugs in clinical practice, our results suggest that Rheumazin can be a good choice for third molar removal treatment.

ABSTRACT

ABSTRACT

ABSTRACT

Marine species muscles present non-proteins nitrogenated compounds, used as quality index. They are total volatile basis (NBVT), trimethylamine oxide (TMAO) and trimethylamine (TMA). pH is considered too as a quality index. The aim of this work was to evaluate these parameters in a fresh and canned marine product from the V region, corresponding to mora crab (Homalaspis plana). Fresh pincer meat from mora crab was extracted and kept in ice until theits analysis and thermal process of the canned product. A 3(2) statistical design was applied, considering two variables with 3 levels: 15, 30 y 45 minutes time levels: 80 degrees, 100 degrees y 121 degrees C temperature levels. Nine conditions of time-temperature were obtained. The thermal treatment caused an increase in pH and BVT. The TMA was increased since reduction of TMAO.
